產(chǎn)品描述：
色霉素A3（Chromomycin A3，CMA3，CAS：7059-24-7），同義名Aburamycin b，由灰色鏈霉菌（Streptomyces griseus）代謝純化所得的一種蒽醌抗生素，具有抗菌、抗真菌和抗腫瘤活性。常用作一種熒光DNA染料，非常適合用來篩查經(jīng)子染色質(zhì)存在的精蛋白缺乏（protamine deficiency）。在二價金屬離子存在的條件下，色霉素A3（CMA3）通過與DNA特異性結(jié)合與可逆作用來阻斷大分子合成。另外，通過結(jié)合到DNA小溝，色霉素A3有效調(diào)控與DNA促旋酶的競爭性抑制，從而顯著影響拓撲異構(gòu)酶II活性。CMA3對非小細胞性肺癌和宮近癌呈現(xiàn)體外細胞毒性，作用于HCC44，A549，ME180和 HeLa細胞的IC50s分別是1，42，60和40nM。CMA3還能通過改善Sp1和Sp3轉(zhuǎn)錄因子結(jié)合來抑制氧化應激和DNA損傷誘導的神經(jīng)損傷。具體內(nèi)活性。

For CMA3 staining, each slide was treated for 20 min with 100µl of CMA3 solution (0.25 mg/ml in McIlvaine buffer, pH 7.0, containing 10 mM MgCl2). The slides were then rinsed in buffer and mounted with buffered glycerol. Microscopic analysis of the slides was performed on a fluorescent Zeiss axioplane microscope with the appropriate filters. |
